盐酸洛拉曲克对移植性肿瘤的生长抑制作用

摘　　要：目的研究盐酸洛拉曲克(Nolatrexed Dihydrochloride,AG337)(化学名Thymidine-110简称THY-110)对小鼠移植性肝癌(H22)、艾氏腹水癌(EAC)和肉瘤(S180)的生长抑制作用。方法体重为(18±2)g小鼠,雌雄各半。设80、120、180mg/kg3个给药剂量组,氟尿嘧啶注射液为阳性对照药,5%葡萄糖溶液为阴性对照。小鼠经左前肢腋下接种0.2ml(1×107/ml)肿瘤细胞悬液,接种后连续腹腔注射给药9d。停药后剥离瘤块称瘤重,计算肿瘤生长抑制率。结果THY-110高剂量组对肝癌(H22)的抑制率为(61.2±2.55)%,艾氏腹水癌(EAC)为(65.7±1.25)%,肉瘤(S180)为(74.0±1.25)%;阳性对照药氟尿嘧啶注射液为(61.8±7.7)%。结论THY-110对肝癌、艾氏腹水癌和肉瘤均具有明显的生长抑制作用。Objective To investigate the inhibition effect against transplatation tumors H22 , EAC and S180 by abdominal cavity injection of THY-110 in mice.Method Mice were divided into five groups including three drug groups i.e. with THY-110 dose of 80, 120 and 180 mg/kg and a negative（glucose injection） group and a positive drug（fluorouracil） group. 0.2 ml（1×10^7/ml）tumor cell solution were inoculated to mice at forelimb oxter. After inoculation,mice were continuous injected of THY-110 in 9 days,and then tumors tissue were peeled off and weighed. The inhibitory rate of tumors were calculated based on tumors weight.Result The average tumor inhibitory rates of THY-110（180 mg/kg） against H22,EAC,S180 were 61.2±2.55%,65.7±1.25%,74.0±1.25% respectively,and that of positive drug group was 61.8±7.7%. Conclusion THY-110 has a distinct effect for inhibiting H22 , EAC and S180 transplation tumors in mice.
