Study on Relativity of the Chondrocytes Apoptosis and the Subchondral Bone Biomechanics in Earlier Period of Osteoarthritis of Rabbit

摘  要:目的探讨骨性关节炎(OA)早期关节软骨细胞凋亡与软骨下骨生物力学改变及其相关性。方法取24只新西兰大白兔,采用Hulth关节不稳造模,左膝为模型组,右膝为对照组,10 d、3个月时各取12只放血处死。各时相点模型组、对照组各取4个膝关节的胫骨近段,去除软组织,进行Mankin软骨损伤评分,采用Tunel法检测关节软骨细胞凋亡情况。去除软骨后用万能力学试验机测定软骨下骨的载荷-位移曲线、应力-应变曲线及弹性模量。结果在造模10 d、3个月后软骨细胞的凋亡逐渐增加,与对照组的差异有高度统计学意义(P<0.01);10 d模型组和3个月模型组的软骨下骨的弹性模量与对照组比较,分别相差18%和19%(P<0.05);关节软骨细胞凋亡数增加与软骨下骨的弹性模量下降变化呈负相关(r=-0.7579,P<0.01)。结论OA早期即出现软骨细胞凋亡,同时软骨下骨生物力学发生改变。Objective To explore the relativity of chondrocytes apoptosis and the subcbondral bone biomechanics in osteoarthritis of rabbit. Methods With Hulth method the left knee of 24 New Zealand white rabbits were divided into the model group and the right knee into the control group. 12 rabbits were killed after 10 days and the others were killed after 3 months. The Mankin score of the 4 rabbits knee with 50 % solution of formic acid and HE staining were made. The chondrocytes apopto- sis of the other 4 rabbits knees were detected with the Tunel method. The stress-strain curves, the load values and the moment of specimens submitting and the elastic modulus were tested in the last 4 rab- bits knees. Results In the model group the chondrocytes apoptosis increase gradually with time. The chondrocytes apoptosis had obviously statistical differences in the two groups(P (0.01). Meanwhile, the elastic modulus had obviously statistical differences in the two groups(P (0.05). The chondro- cytes apoptosis were in negative correlation with the elastic modulus of subchondral bone (r = - 0. 7579, P (0.01). Conclusion In earlier period of OA the chondrocytes apoptosis is emerged and the cartilage biomechanics is changed.
