摘  要:目的探索脑源性神经生长因子(BDNF)在诱导骨髓间充质干细胞(BMSCs)分化为神经元样细胞过程中的作用。方法应用Ficoll分离骨髓中单核细胞,贴壁培养细胞,观测形态并用流式细胞仪检测其细胞表面标志;用含β-巯基乙醇(BME)、BDNF及碱性成纤维细胞因子(bFGF)的条件培养基使培养的细胞向神经细胞诱导分化;采用免疫荧光染色法对分化的细胞进行鉴定。结果分离培养的贴壁细胞具有典型的BMSCs的形态和细胞表面标志,诱导后细胞表达神经元和星型胶质细胞标志神经元特异性烯醇化酶(NSE)、和胶质纤维酸性蛋白(GFAP)。结论BMSCs在体外具有分化为神经元样细胞的能力,BDNF具有促进分化的作用。Objective The aim of this study is to search for the effect of brain-derived neurotrophic factor(BDNF) during inducing bone-marrow mesenchymal stem cells (BMSCs)into neurons-like cell in vitro. Methods First, monocytes from bone marrow with Ficoll and these cells were incubated by adherent culture. Then were these cells morphology and surface markers examined. Finally the culture cells were induced to differetiate into neuron-like cells by β-mercaptoethanol(BME), BDNF and basic fibroblast growth factor (bFGF), the differentiated cells were identified with immunofluorescence. Results The culture cells had the typical morphology and cell surface antigen of BMSCs and could be induced to differetiate into neuron-like cells, exhibit the neuonral phenotype, express neuron specificenolase(NSE) and glial fibrillary acid protien(GFAP). Conclusion BDNF can promote the differentiation of BMSCs into neuron-like cells.
