摘 要:目的研究氡暴露致小鼠骨髓细胞的氧化损伤。方法采用雄性BALB/C小鼠24只,随机分为4组,每组6只,除对照组外,处理组小鼠整体暴露于多功能生态氡室,吸入氡及其子体的累积剂量分别为27(低剂量组)、52(中剂量组)、105(高剂量组)工作水平月(working level month,WLM)。测定骨髓细胞的超氧自由基(O2-)浓度、氚-胸腺嘧啶核苷(3H-TdR)掺入能力、超氧化物歧化酶(SOD)活性和脂质过氧化物(LPO)的含量。结果各处理组骨髓细胞的O2-浓度均较对照组明显上升,差异均有统计学意义(P<0.05)。而骨髓细胞的增殖能力即cpm值有不同的变化,其中52 WLM组与对照组、27 WLM组相比均明显增加,而105 WLM组与对照组、52 WLM组相比则明显降低,差异均有统计学意义(P<0.05)。与对照组相比,各处理组SOD活性均有显著性降低,LPO含量均明显上升,差异均有统计学意义(P<0.05),其中105 WLM组的变化更为显著。结论高剂量氡暴露可对小鼠骨髓细胞产生氧化损伤,并抑制骨髓细胞的DNA合成。Objective To study toxic effect of radon exposure on hone marrow cells of mice to explore mechanisms of radon-induced leukemia. Methods Twenty-four mice were randomly divided into four groups, one control group and three experimental groups with the cumulative doses of radon at 27 WLM(low dose group), 54 WLM(middle dose group) and 105 WLM(high dose group).Levels of superoxide dismutase (SOD), lipid peroxidation (LPO), ^3H-TdR incorporation and superoxide free radical in bone marrow cells were measured to reflect the lipid peroxidation induced by radon. Results The level of superoxide free radicals of the control group shows marked increase compared to other three experiment groups, the difference between them all have statistical significance (P〈0.05). But reproduction ability of marrow cells, namely cpm shows different change trend. The cpm value of 52 WLM group is higher than that of the control group and 27 WLM group, but the cpm value of 105 WLM group is lower than that of the control group and 52 WLM group (P〈0.05). Apparent decrease of SOD of all experiment groups are observed compared to the control group, but as for LPO, contrary situation appear (P〈0.05). The relative changes Of 105 WLM group is most obvious. Conclusion Damages of oxidation and DNA synthesis are observed in bone marrow cells in the high dose group.
