摘  要:随着腔前卵泡体外培养体系的发展,对其影响因子的研究也逐渐深入,促性腺激素(FSH)在卵泡的生长和发育过程中发挥了重要的作用。本实验方法是以昆明小鼠为模型,机械分离并选择120~140μm的腔前卵泡,以添加10%血清和1%ITS的α-MEM为基础培养,分为正常添加FSH和不添加FSH组,以及在培养的0d、2d、4d、6d、8d添加FSH(100mIU/m1)组,探讨腔前卵泡的生长发育情况。结果表明:正常添加组其卵泡的存活率、出腔率、GVBD率和2-cell胚胎率(78.7%、55,0%、35.0%和7.5%)显著高于培养液中未添加FSH的结果(分别为13.7%、8.8%、6.3%和0)(P〈0.05);6d之前添加FSH的培养组腔前卵泡能够正常生长和发育;2~4d添加FSH可能更利于卵母细胞的成熟,以及E2的产生依赖于FSH的存在。Along with the progress of culture system on preantral follicles, the study became deeply in influential factors. Follicle stimulation hormone (FSH) has been demonstrated to play an important role in growth and development of follicles, but it is unknown if FSH can act on the growth of preantral follicles. In this paper, 120 - 140μm preantral follicles were taken from mice and being cultured in the medium consisted of a-minimal essential medium (α-MEM) , supplemented with FCS and ITS. 100 mIU/ml FSH was introduced from day 0, 2, 4, 6 or 8 during culture. No FSH was contained in control group. The results showed that the survival rate, antral-like cavity rate, GVBD rate and 2-cell rate in group cultured with FSH continuously(78.7% , 55.0% , 35.0% and 7.5% respectively)were higher than the group that cultured without FSH (13.7%, 8.8%, 6.3% and 0% respectively)(P 〈0.05). Although the preantral follicles grew and developed normally when FSH was added at the sixth day, it seems that addition of FSH from the second to the fourth day maybe more feasible for the development of oocytes. Moreover, FSH supplement would enforce Estradiol hormone production.
