霍乱弧菌O139和ElTor菌株毒力岛区域分子特征比较

摘　　要：目的:比较O139霍乱弧菌(VCO139)菌株MO45基因组与O1群ElTor霍乱弧菌(EVC)菌株N16961基因组中毒力岛(VPI)区域分子特征。方法:制备霍乱弧菌VPI上游VC0815基因探针。从VCO139菌株MO45基因组文库中筛选一个既含有VC0815基因又含有tcpA基因的克隆,命名为pCOSVC081545,绘制pCOSVC081545VPI上游片段的限制性酶切图谱,并与N16961同段序列的限制性酶切图谱进行比较。结果与结论:pCOSVC081545和N16961中VPI上游约1500bp片段的限制性酶切图谱基本相同,支持O139霍乱弧菌是EVC O抗原突变而来的观点。Aim： To compare molecular character of Vibrio cholerae pathogenicity island （VPI） of Vibrio cholerae O139 （VC O139） MO45 and E1 Tor biotype of Vibrio eholerae （EVC）N16961. Methods：A probe of VC0815 that lay in the upriver of VPI was prepared. The clones that including VC0815 and tcpA genes were screened from the genome libraries of MO45 and named pCOSVC081545. The restriction map of the upriver segment of VPI of pCOSVC081545 was drawn and compared with that of the same segment of N16961. Results and Conc（usion ： The restriction maps of the upriver segment of VPI of pCOSVC081545 were similar to that of N16961 ,which suggested that VC 0139 maybe result from the mutation of the O antigen of EVC.
