摘  要:目的:观察淫羊藿总黄酮注射液(EFI)对犬急性实验性心肌梗死的治疗作用及对H_2O_2诱导乳鼠心肌细胞凋亡的影响。方法:采用左冠状动脉前降支(LAD)两步结扎复制犬急性心肌梗死模型,心肌切片氯化硝基四氮唑蓝(N-BT)染色计算心肌梗死范围,测定血清游离脂肪酸(FFA)和乳酸(LA)水平。通过体外培养大鼠乳鼠心肌细胞,流式细胞仪和琼脂糖凝胶电泳检测心肌细胞凋亡。结果:5,10和15 mg·kg^(-1)EFI均能显著缩小犬心肌梗死范围,降低血清FFA和LA水平。100μmol·L^(-1)H_2O_2作用12h后乳鼠心肌细胞凋亡率显著高于正常对照组,且出现典型凋亡的DNA梯带;100,200和400 mg·L^(-1)EFI可剂量依赖性降低乳鼠心肌细胞凋亡率,同时可减少DNA梯带形成。结论:EFI对犬急性实验性心肌梗死有较好的治疗作用,对H_2O_2诱导的乳鼠心肌细胞凋亡具有保护作用。Objective: To observe the protective effect of epimedium flavonoids injection (EFI) on acute myocardial infarction (AMI) in dogs, and on H2O2 -induced apoptosis in neonatal rat cardiomyocytes. Methods: AMI was induced by ligating the left anterior descending coronary artery (LAD) in anaesthetized dogs. The myocardial infarct size was determined by nitroblue tetrazolium (N-BT) staining. The levels of serum FFA and LA were measured. In the cultured ventricular myocytes isolated from neonatal rats, apoptosis was determined by flow cytometry after propidium iodide (PI) staining and by agarose gel electrophoresis of DNA fragmentation. Results: In the AMI dogs, EFI at 5, 10 and 15 mg·kg^-1 decreased the infarction size, and the levels of serum FFA and LA. In the neonatal rat cardiomyocytes, apoptosis rate was significantly increased and typical DNA ladder appeared after exposure to 100 μmol·L^-1 H2O2 for 12 h. EFI at 100, 200 and 400 mg·L^-1 concetration-dependently decreased the apoptosis rate and DNA ladder. Conclusion: EFI has a protective effect on acute myocardial ischemic infarction in dogs and on H2O2-induced apoptosis in neonatal rat cardiomyocytes.
