摘 要:目的:探讨桦褐孔菌提取物对肿瘤细胞周期的影响及其抗癌的作用机制。方法:将不同浓度(20、40、80mg·L-1)桦褐孔菌提取物作用于人胃癌BGC-823细胞48h,用碘化丙啶染色法检测凋亡细胞和细胞周期分布。采用免疫细胞化学染色法检测药物处理前、后Ki-67抗原的表达。结果:3种浓度桦褐孔菌提取物处理48h后细胞凋亡率分别为11.57%、20.97%、31.22%,阻滞细胞周期于S期,并使Ki-67阳性表达率下降,且具有浓度依赖性。结论:桦褐孔菌提取物可抑制人胃癌BGC-823细胞的生长,细胞主要阻滞于S期,并能诱导部分细胞凋亡。OBJECTIVE: To investigate the possible mechanism of anticancer activity of the extracts from Inonotus obliquus and its association with the phase of cell cycle, METHODS: FCM was used to detect cell cycle distribution of human gastric carcinoma BGC- 823 cells that treated with extracts from Inonotus obliquus at different concentrations (20, 40, and 80 mg· L^-1) for 48h and their apoptotic rate, marked by PI. Furthermore, the protein expressions of Ki- 67 were detected by immunocytochemical staining method before and after exposure to extracts from Inonotus obliquus. RESULTS: After being treated by extracts from Inonotus obliquus (20, 40, and 80mg · L^-1) for 48h, the percentage of apoptosis cells were 11.57%, 20.97%, 31.22%, respectively and the human gastric carcinoma BGC-823 cells were inhibited and arrested in S phase. The protein expression of Ki- 67 was decreased dose- dependently. CONCLUSION: The extracts from Inonotus obliquus can inhibit the growth of human gastric carcinoma BGC - 823 cells much as in S phase and induce apoptosis of BGC-823 cells.
