摘  要:目的探讨赤芍总苷(TPG)静脉给药抗血栓作用以及其作用机制。方法通过动静脉旁路血栓形成实验,观察TPG对血栓形成的抑制作用;建立电刺激致大鼠血栓模型,观察TPG抗血栓作用以及对血液流变学指标、凝血-纤溶系统的影响;TPG静脉注射给药5d后,测定其对小鼠凝血时间的影响;以体外法观察TPG对ADP诱导家兔血小板聚集的影响。结果TPG25、50、100mg/kg能明显抑制大鼠动-静脉旁路血栓的形成,抑制率分别为32.59%、34.33%、24.88%;TPG25、50、100mg/kg能明显延长电刺激大鼠动脉血栓形成所需的时间,所需时间分别为648.2s、651.1s、746.4s,与对照组(540.2s)比较,差异有统计学意义(P<0.05);降低血粘度,延长凝血酶原时间和部分凝血活酶时间,降低血纤维蛋白原浓度;有效延长小鼠的凝血时间;显著抑制ADP诱导的血小板聚集(抑制率分别为23.4%、37.2%、44.9%)。结论TPG具有较好的抗血栓作用,其作用机制与其能降低血粘度,延长大鼠凝血酶原时间、部分凝血活酶时间以及降低血纤维蛋白原浓度,抑制血小板聚集有着密切的关系。Objective To study the antithrombotic action and mechanism of total paeony glucoside (TPG) Methods The arterio-venous shunt thrombosis model was used for this study. The thrombosis model was established by electrical stimulus in rats. TPG was injected intravenously. The effects of TPG on blood rheology, fibrolysis, blood clotting and platelet aggregation induced by ADP were observed after 5 days of injection. Results TPG can obviously inhibit the thrombosis made by arteriovenous shunt and electrical stimulus, lower the blood viscosity, extend the prothrombin time and slightly extend the thromboplastin time, lower the fibrinogen concentration; extend the clotting time ; and inhibit the platelet aggregation induced by ADP. Conclusion TPG has anti-thrombosis effect. The mechanism of action may be due to TPG can lower the blood viscosity, extend the prothrombin time and slightly extend thromboplastin time, lower the fibrinogen concentration; and inhibit the platelet aggregation.
