摘  要:将人精液分为两组,一组直接用于制备精子单倍染色体,一组保存于4~8℃的HT卵黄缓冲液中48小时,再用于制备精子单倍染色体。比较两组的可分析染色体获得率与自然畸变率,了解HT卵黄缓冲液是否会造成自然畸变率的上升和可分析染色体获得率的下降。对35份样品试验与观察得到两组间可分析染色体获得率比较结果,X2=0.159,P>0.05;自然畸变率两组比较结果,X2=0.074,P>0.05。结果表明两组间在这两个率比较中无统计学差异,HT卵黄缓冲液可应用于精子在冷环境中短期贮藏,并不具有致染色体突变的效应。Human semens were separated into two groups. The one group semen was used to preparing the sperm haploid chromosome directly. Another group semen were stored in HT yolk buffer for 48 hours at 4~8℃,then the sperms from the semen were resuscited at 37℃ for preparing the sperm haploid chromosome. During 35 times repeating experiments and comparing the rates of analysible sperm chromosome and nature distortion of the chromosome between the two groups, there are not any statistical differences between the two groups' rates ( P >0.05). The HT yolk buffer does not have any effect of making the chromosome distortion. The HT yolk buffer can be used in storing human sperm at 4~8℃ for a short term for clinic examination and some research purpose in andrology.
