烟酸对高脂血症兔主动脉斑块面积和MCP-1的影响

摘  要:【目的】观察烟酸对动脉粥样硬化斑块形成及单核细胞趋化因子-1(MCP-1)的影响。【方法】将新西兰兔给予高胆固醇饮食8周后随机分为高脂组、烟酸组和对照组。第14周末处死动物进行主动脉病理学检测,酶联免疫吸附法测定MCP-1浓度和半定量逆转录聚合酶链反应测定MCP-1 mRNA的表达。【结果】高胆固醇饮食14周后,兔主动脉内膜较对照组显著增厚,并明显可见斑块形成。烟酸干预6周后胆固醇水平显著低于高脂组(P<0.05);主动脉内膜厚度小于高脂组,斑块面积也显著小于高脂组(P<0.01)。正常饮食兔的MCP-1水平很低[(61.4±10.2)pg/mL],高胆固醇饮食后显著升高[(227.1±23.0)pg/mL],烟酸干预6周后则显著降低[(164.5±21.7)pg/mL,P<0.01];血清MCP-1水平与动脉粥样硬化斑块的面积呈正相关(r=0.71,P<0.01)。【结论】烟酸延缓动脉粥样硬化斑块的形成作用可能与其降脂及降低MCP-1水平有关。[Objective]To investigate the effects of niacin on the plaques formation of aorta and MCP-1 expression in hypercholesterolemic rabbits, [Methods]Twelve male New Zealand white rabbits were fed with high-cholesterol diet for 8 weeks , and then were randomly divided into two groups: (1)high cholesterol group ( n =6): maintained on high cholesterol diet for 6 weeks; (2)niacin group ( n =6); the same cholesterol diet plus niacin [200 mg/(kg·d)] for 6 weeks. Control group ( n =6) was fed with normal diet for 14 weeks. Serum MCP-1 concentrations were detected by enzyme-link immunosorbent assay, and the histopathological changes of the aortas were detected at the end of the study. MCP-1 mRNA expression in aortas was evaluated by reverse transcription-polymerase chain reaction (RT-PCR). [Results]Compared with control group, rabbits fed with high cholesterol diet showed higher levels of serum total cholesterol, LDL cholesterol and MCP-1, all of which were significantly reduced by niacin treatment. There were atherosclerotic lesions in aortas in high cholesterol group as compared with control group. The areas of the plaque [ (45.7±8.9) %] and the intima thickness [(28.54±6.37)μm] of the aorta were ameliorated in niacin group as compared with high cholesterol group [(81.6±10.2)%, (75.36±11.25) μm, respectively]. The base concentrations of MCP-1 in the three groups were very low. The levels of MCP-1 were significantly increased after 8 weeks of high cholesterbl diet in starch group, and niacin significantly reduced the level of MCP-1 [(164.5±21.7) pg/mL] as compared with high cholesterol group [(227.1±23.0) pg/mL] at the end of the study. The concentration of MCP-1 was positively associated with the plaques areas of aorta (r = 0. 71, P〈0.01). [Conclusion]Niacin can decreased the serum cholesterol, MCP-1 concentrations and aorta MCP-1 mRNA expression, which may inhibit the formation of plaques of aortas in hypercholesterolemic rabbits.
