摘　　要：目的筛选蓝氏贾第鞭毛虫的保护性抗原,为免疫疫苗的研制奠定基础。方法利用已建立的蓝氏贾第鞭毛虫滋养体体外培养系统,获取滋养体抗原,并进一步分离、纯化、活性鉴定;然后分组免疫小鼠,检测相关免疫指标;再行攻击感染试验,视其抗感染能力,筛选出保护性强的抗原组分。结果成功获取5个组分的抗原蛋白(P76/66、P54/50、P34/32、P30/29和P21/19),P76/66抗原活性最强,其诱发BALB/C小鼠产生的抗体含量最高。各组分抗原免疫小鼠,产生的白细胞介素-2(IL-2)、γ干扰素(IFN-γ)及CD4+、CD8+T淋巴细胞的数量均随免疫次数的增多而逐渐增加。攻击试验结果显示,P76/66抗原的免疫保护率最高为100%,其次是P21/19为65%。结论P76/66抗原具有良好的免疫保护力,为蓝氏贾第鞭毛虫免疫疫苗的研制提供了物质基础。Objective To screen the protective antigen of G. intestinalis and set the foundation for immune rescach of vaccine. Methods The trophozoite antigens were obtained with in vitro culture system of Giardia lamblia Stile. After further separation, purification and activity verification, the antigens were used to immunize the BALB/C mice divided into different groups, then indexes associated with immunity were determined. Through challenge infection, the most protective antigenic component of G. intestinalis were selected based on its anti - infection ability. Results There 5 components of antigenic albumen （P76/66,P54/50,P34/32, P30/29和 P21/19） were successfully obtained, among them the P76/66 antigen activity was the strongest, which induced the maximum level of antibody in the mice. The levels of interleukin - 2 （IL - 2） γ/ interferon （IFN -γ） and CD4^＋ ,CD8^＋ T lymphocyte would be increased along with the increase of times of immunization in the mice in each group. The results of challenge infection revealed that the highest protecting rate of immunity of P76/66 antigen was 100%, followed by P21/19 （65%）. Conclusion P76/66 antigen has a satisfactory ability of inducing an immune protection in mice and provides the material basis for the development of vaccine against G. intestinalis .
