摘 要:【目的】探讨罗格列酮(RSG)对血管损伤后内膜增生的影响及可能机制。【方法】制备大鼠胸主动脉球囊损伤模型,将SD大鼠随机分为RSG组、对照组和假手术组,于术后d14处死动物。分别进行伊文思蓝染色,细胞核增殖抗原(PCNA)、P27免疫组化染色和组织形态学定量分析,并用流式细胞术测定各组平滑肌细胞各期(G0/G1期,S期,G2/M期)的百分比。【结果】RSG组的再生内皮覆盖率较对照组显著增加(P<0.01),且该组动脉内膜面积与中膜面积的比值(IA/MA)较对照组显著降低(P<0.01)。与对照组相比,RSG组新生内膜内的PCNA性表达指数均显著减少(P<0.01),而P27的表达显著增强(P<0.01),流式细胞术结果显示平滑肌细胞处于增殖期的细胞比例明显降低(P<0.05)。【结论】RSG可以明显抑制动脉损伤后的内膜再生,其机制可能与上调P27及细胞周期调控有关,此外RSG还可能通过促进损伤处内皮再生来抑制内膜增生。[Objective]To investigate the effects of rosiglitazone(RSG) on neointimal formation after balloon injury and its mechanism. [Methods] Endothelial denudation of aorta in SD rats was performed with 2F Fogarty balloon catheter. Male rats were randomly divided into sham-operated group, control group, and RSG group. Aortic tissues were harvested in 14th day after injury. The endothelial regeneration rate, IA/MA ratio,P27 and PCNA expression index , and cell cycles were detected. [Results] Re-endothelialization of the injured aorta in RSG group was enhanced as determined by Evans blue stain( P 〈0.01). There was a significant decrease of the neointima in RSG groups( P 〈0.01). The expression of P27 was greater and that of PCNA was lower in RSG group than that in control group( P 〈0.01). The Flow Cytometry showed that the percentage of S and G2/M phase cells in RSG group were lower than that in control group. [Conclusion] RSG can obviously attenuate neointimal formation of the aorta after balloon injury and its mechanism may be related to upregulation of P27 and inhibition of cell cycle progression . Otherwise RSG may also inhibit proliferation of the intima by promoting endothelial regeneration.
