地塞米松对急性肺损伤大鼠水通道蛋白1表达的影响

摘  要:目的研究急性肺损伤(ALI)大鼠肺组织中水通道蛋白1(AQP1)的表达变化,探讨地塞米松治疗ALI的可能机制,为临床治疗ALI开辟新的治疗措施提供理论依据和实践方向。方法30只SD大鼠随机分成3组(每组10只),即油酸致ALI组(OA组)、地塞米松治疗组和生理盐水对照组(NS组)。观察6 h后测定动脉血氧分压(PaO2)、二氧化碳分压(PaCO2)、pH值、肺湿/干(W/D)比值,并采用免疫组化方法检测各组大鼠肺组织中AQP1的表达变化及对肺组织进行常规病理染色。结果与NS组相比OA组AQP1的表达显著降低(P〈0.05),而地塞米松疗组AQP1的表达显著高于OA组(P〈0.05),同时地塞米松治疗组能使血气明显改善(P〈0.05),W/D比值下降(P〈0.05),肺组织损伤减轻。结论地塞米松对ALI大鼠AQP1表达的调节可能是其治疗ALI的有效途径之一。Objective To explore the mechanism of dexamethasone in the treatment of acute lung injury(ALI) in rats induced by oleic acid(OA) by studying changes of expression of aquaporin 1(AQP1),so providing a new clinical method in treating ALI.Methods Thirty Sprague-Dawley rats were randomly divided into three groups which were ALI induced by OA injection(OA group,n=10),therapy group treated by dexamethasone(Dex group,n=10) and normal control treated by saline injection(NS group,n=10).Arterial oxygen tension(PaO2),arterial carbon dioxide tension(PaCO2),power of hydrogen(pH) and wet/dry mass ratio(W/D) were assayed at six hours.Changes of AQP1 expression in the lungs of rats were analyzed by immunohistochemistry and pathological examination were performed in each group.Results Compared with NS group,AQP1 expression in OA group decreased significantly(P〈0.05),while AQP1 expression in Dex group was higher than that in OA group(P〈0.05).Dex group could improve the blood air level(P〈0.05),reduce the W/D mass ratio(P〈0.05) and relieve the lung injury.Conclusions Dexamethasone may have beneficial effects on ALI by regulating AQP1 expression in the lungs of rats.
