摘  要:目的:研究经前平颗粒对经前期综合征肝气逆证模型大鼠下丘脑中雌二醇α受体和孕酮受体表达的影响。方法:以情志刺激多因素法复制经前期综合征肝气逆证大鼠模型,经前平颗粒预防给药(10 g.kg-1,灌胃给药,连续5 d),采用免疫荧光细胞化学技术结合流式细胞仪检测大鼠下丘脑中雌二醇α受体和孕酮受体的蛋白表达量。结果:模型组大鼠下丘脑中雌二醇α受体和孕酮受体蛋白表达量与正常组相比显著降低(P<0.01),经前平颗粒组下丘脑中雌二醇α受体和孕酮受体蛋白表达量较模型组显著升高,接近于正常水平。结论:经前平颗粒可以上调下丘脑雌二醇α受体和孕酮受体蛋白表达水平,这可能是其治疗经前期综合征肝气逆证的作用机制之一。Objective: To study the effects of Jingqianping granule on the expression levels of estradiol receptor alpha and progesterone receptor in hypothalamus of premenstrual syndrome (PMS) model rats with liver-qi invasion. Methods: The PMS model with liver-qi invasion was induced in rats by multiple emotional stimulations. Jingqianping granule (10 g·kg^-1, ig) was administered for 5 d before and after PMS induction. The expression levels of estradiol receptor alpha and progesterone receptor in the hypothalamus were detected by fluorescent immunocytochemistry and flow cytometry. Results: The expression levels in hypothalamus of estradiol receptor alpha and progesterone receptor were significantly lower in PMS model rats than in control rats (P 〈 0.01 ). Jingqianping granule markedly increased the reduced expression levels of both receptors, and recovered them near the normal level. Conclusion: Jingqianping granule can up-regulate the expression levels of estradiol receptor alpha and progesterone receptor in hypothalamus after PMS, which may be one of the mechanisms underlying the effects on PMS with liver-qi invasion.
