口服Ⅱ型胶原及抑肽酶对佐剂型关节炎大鼠治疗作用研究

摘　　要：目的研究口服Ⅱ型胶原(CⅡ)及抑肽酶对大鼠佐剂型关节炎的治疗作用。方法将50只大鼠随机分为5组,采用完全佐剂制备大鼠佐剂型关节炎模型。按不同方案口服给与CⅡ及抑肽酶后考察对踝关节肿胀的影响。采用免疫荧光技术,用流式细胞仪检测大鼠外周血T细胞亚群的变化,并且用大鼠TNFa检测试剂盒检测大鼠外周血TNFa水平的变化。结果建立了大鼠佐剂型关节炎实验动物模型,Ⅱ型胶原组及Ⅱ型胶原和抑肽酶组与生理盐水组比较均可显著减轻踝关节肿胀的程度,且两组大鼠外周血CD3 T细胞、CD4 T细胞、CD8 T细胞及外周血TNFa水平与生理盐水组比较也均有显著性差异;Ⅱ型胶原组与Ⅱ型胶原和抑肽酶组组间作用比较,后者作用好于前者,部分结果有显著性差异;抑肽酶组与生理盐水组比较,治疗作用差异不显著。结论Ⅱ型胶原中加入抑肽酶后能更有效地治疗大鼠佐剂型关节炎。Objective： To study the treatment effect of type Ⅱ collagen and aprotinin on adjuvant induced arthritis in rat after oral administration. Methods： 50 rats were divided into 5 groups, and the model of adjuvant induced arthritis was prepared by Freund＇ s complete adjuvant. We observed the influence of C Ⅱ and aprotinin in oral administration according to different schema on ankle joint swelling. With immunnfluorescence technique, T cell subsets and TNFa concentration in peripheral blood were assayed by flow cytometer and rat TNFa ELISA kit separately. Results： Adjuvant induced arthritis model in rat was prepared. Compared with group saline, both group C Ⅱ and group C Ⅱ with aprotinin could ameliorate ankle joint swelling of rat, and the T cell of CD3 , CD4 , CD8 and TNFa concentration also had significant difference. Compared between group C Ⅱ and group C Ⅱ with aprotinin, the action of latter was more effective and some results had significant difference, but the treatment effect of group aprotinin had no difference compared with group saline. Conclusion ： C Ⅱ could more effectively cure adjuvant induced arthritis in rat by added aprotinin.
