摘 要:研究了利用蒸发溶剂法、反溶剂法和超临界流体技术将阿维菌素包埋到空心多孔纳米SiO2载体中的工艺过程,并利用热分析仪、智能溶出仪、紫外分光-光度计等对其整体载药量、载体内、外载药量分布以及载药后的缓释性能进行了分析。结果表明:用超临界流体包埋法制备的阿维菌素纳米控释剂(Av-PHSN)的整体载药量和内部载药量均为最大,缓释效果最好。反溶剂法和蒸发溶剂法制备的Av-PHSN载药量虽然不如超临界包埋法高,但工艺流程简单,能耗小,在最优操作条件下也能获得较满意的整体载药量和载体内部载药量。The loading of avermectin into porous hollow silica nano-particles (PHSN) by the methods of vaporizing solvent (VS), anti-solvent precipitation (ASP) and supercritical fluid drug loading (SFDL) has been investigated. The total loaded drug (TD), the drug loaded inside the carrier (DI), the drug loaded outside the carrier (DO) and the sustained release behavior of the avemectin-loaded PHSN samples were analyzed by thermogravimetry (TG), UV spectroscopy and dissolution testing. The results indicated that the highest values of TD and DI and best sustained release behavior were achieved using the SFDL method, but this method also has the highest operating cost. Under the optimal operation conditions, VS and ASP methods give a moderate drug loading, and therefore, in cases when very high TD and DI are not required, VS an.d ASP methods can be a good alternative for SFDL due to their simpler and lower cost operation.
