摘 要:以常用的植物基因组DNA提取方法—LiCl法、SDS法、CTAB法、高盐脲法和试剂盒法提取条斑紫菜叶状体DNA,对不同提取方法的效果进行了比较分析,同时探讨了SDS法的细胞破碎方法和不同样本量提取条斑紫菜DNA的效果。对DNA纯度、DNA浓度、DNA得率等分析表明,SDS法效果较好,其次是LiCl法;在使用SDS法时,液氮研磨破碎细胞效果好于其他方法;在小样本量提取DNA时,SDS法比LiCl法效果好。DNA from thallus of Porphyra yezoensis was extracted by method of LiCI, SDS, CTAB, Highsalt Urea and Plant Genomic DNA Isolation Kit, and the effects of different extraction methods were compared and analyzed. At the same time, the effects of smudge cell methods and differrent sample sizes to extract the genomic DNA of Porphyra yezoensis were discussed. By evaluating purification of DNA, concentration of DNA, and DNA yield, the results indicated that the effect of SDS was better than other methods, and then was LiCl. When SDS was used, the smudge cell by skive with liquid nitrogen had the best effect among these methods. SDS to extract DNA was better than LiCl in small sample.
