摘  要:研究辐射对肝癌细胞中乏氧诱导因子-1α(HIF-1α)的调控作用。采用氯化钴(CoCl2)化学模拟乏氧,诱导HepG2肝癌细胞内HIF-1α稳定表达,再进行不同剂量的γ射线照射,观察照射后细胞内HIF-1α表达的变化,同时,利用荧光显微镜和流式细胞术(flow cytometry,FCM)对细胞内的活性氧(reactive oxygen species,ROS)含量进行检测。结果显示,在乏氧条件下HepG2细胞受1―5Gyγ射线照射后,细胞内HIF-1α的表达与辐照剂量呈正相关。在1―3Gyγ射线照射后,细胞内活性氧的含量与HIF-1α的表达呈负相关。此研究提示,辐射可增强乏氧细胞内HIF-1α的表达水平,活性氧含量的降低可能与这一调控作用有关。This paper reports regulation of hypoxia inducible factor-1 α (HIF-1 α) in hepatoma cells by irradiation. Cobalt chloride (CoCl2), a chemical mimic agent for hypoxia research, was utilized to induce the stable expression of HIF-1α in HepG2 cells. The HepG2 cells were irradiated to different doses to observe the changes of HIF-1α. The level of intracellular reactive oxygen species (ROS) was assayed by fluorescent microscope and flow cytometry (FCM). The results showed that there were obvious changes in expression of HIF-1α after HepG2 cells exposed to radiation, and the changes were positively related with the irradiation dose from 1Gy to 5Gy. Moreover, contents of incellular ROS were negatively correlated with above levels of HIF-1α from 1Gy to 3Gy. The results indicate that irradiation may enhance hypoxic cells HIF-1α , and the reduction of intracellular ROS can contribute to the regulation of ionizing radiation on HIF-1α.
