四倍体盾叶薯蓣实生苗的诱导及快速繁殖研究

摘　　要：盾叶薯蓣(D.zingiberensisC.H.Wright)是我国特有的药用植物,根茎所含薯蓣皂苷元为合成甾体激素类药物的主要原料。采用秋水仙碱溶液诱导盾叶薯蓣种子,种子经诱导出芽率明显下降;出芽后所得实生苗采用根尖染色体、花药壁细胞染色体和花粉母细胞染色体计数相结合的方法进行突变体的检测,突破了根尖染色体计数试验的取材限制;经诱导得到了盾叶薯蓣同源四倍体及非整倍实生苗;四倍体单株小孢子母细胞减数分裂行为正常,纺锤丝可见。试验还对四倍体盾叶薯蓣组织培养丛生芽诱导和生根培养基进行了优选。D.zingiberensis C.H.Wright is a kind of endemic plant only growing in China.Diosgenin beening in rhizomes is the main material of to synthesize steroid.In this paper aberrance of D.zingiberensis C.H.Wright was induced by colchicime and then the seed germination rate of D.zingiberensis C.H.Wright was reduced.And the tetraploid were obtained by means of counting the number of chromosome with root,anther and microspore as materials.The meiosis of microspore of tetraploid was in gear.And the optional tissue culture medium was selected in this paper.
