摘 要:给怀孕第三天小鼠注射羟泰米粉,可使子宫雌激素受体基因(ERmRNA)表达明显改变,与给药前比较,6和12小时组ERmRNA显著减少,与不同时间各自的对照组比较,这二个时间组的数值也显著降低;同时还观察到24小时组ERmRNA明显增加,与以往分析雌激素受体(ER)蛋白水平的结果比较,可以看出ERmRNA与胞浆ER的动态变化具有一致性。Previous studies in our laboratory indicated that estrogen antagonist hydroxytamoxifen(OHT)can markedly block embryo implantation in mice during the early pregnant stage(D3).These inhibitory effects on implantation may be mediated by uterine estrogen receptor(ER),because antiestrogen caused changes in the content of estrogen and progesteron receptor (PR) by D3 treatment with 50μg of OHT in ethanol/saline.In order to understand whether this effect of OHT on estrogen receptor occurs at the ERmRNA transcription level,the changes of ER in mouse uterus were assayed by Northern blotting after treatment of the animal with OHT on D3 of pregnant.The results were as follows,ERmRNA slightly increased at 1 hour after antiestrogen treatment and lowered at 2 hour,but there was no significance as compared with those of control animals before injection.At 6 and 12 hours the values of ERmRNA decreased remarkably to 35 32% and 27 1% of zero hour value,respectively.At 24 hour ERmRNA increased greatly,approaching the zero hour value. The values of ERmRNA in OHT group were 43 91 and 33 33% of their respective control values at 6 and 12 hours and went up to 312 24% of its control value at 24 hour.These results showed clearly that ER gene expression in mouse uterus during preimplantation were strikingly changed by OHT administration.Changes in the level of ERmRNA were consistent with those of cytosol ER(ERc) determined formerly.This proved that effect of OHT on ER occurred at the transcription step of ER gene expression.
