摘 要:目的探讨牛磺酸锌(TZC)对汞染毒大鼠海马胆囊收缩素(CCK)神经元的保护作用及其与动物学习记忆功能变化的关系。方法选用Wistar大鼠32只,分为对照组、汞染毒组(每日饮用4.3 mg/kg氯化汞水溶液)、汞+低TZC组(汞染毒的同时饮0.23 g/L TZC水)和汞+高TZC组(汞染毒的同时饮0.46 g/L TZC水)。利用反映学习记忆功能的Y迷宫法测试大鼠神经行为的改变;采用ABC免疫组化检测大鼠海马不同亚区CCK阳性神经元的变化。结果汞染毒组大鼠学习记忆能力明显低于对照组和高TZC组,差异有统计学意义(P<0.05),而高TZC组和对照组的差异无统计学意义(P>0.05)。免疫组化结果显示,汞染毒组大鼠海马CA1区和CA3区CCK阳性神经元数目明显少于高TZC组和对照组,差异有统计学意义(P<0.05)。结论汞可损伤大鼠学习记忆能力和影响海马各区CCK阳性神经元的数目。TZC对汞引起大鼠的学习记忆损伤和海马CCK阳性神经元的变化有保护作用。Objective To investigate the relationship between the effect of taurine zinc coordination compound(TZC) on hippocampal cholecystokinin(CCK)positive neurons and learning and memory ability of chronic mercury( HgCl2 )-exposed rats. Methods Thirty-two Wistar rats were randomly divided into control group, Hg-exposed group, and HgCl2 + low TZC group and HgCl2 + high TZC group. Y-maze test was used to study learning and memory ability of rats; ABC immunohistochemistry and quantitative graphic analysis were used to investigate the changes of CCK positive neurons in different hippocampal subfields of Hg- exposed rats. Results The learning and memory ability in Hg-exposed rats was significantly lower(P〈0.05) than those in control and HgCl2 + high TZC groups. The number and optical density of CCK positive neurons in CA1 and CA3 areas of Hg-exposed rats were significantly lower(P〈0.05)than those in control and HgCl2 +high TZC groups. Conclusions Hg may damage the learning and memory ability and affect the number of CCK positive neurons in Hg-exposed rats. TZC might play an important role in preventing Hg-induced damages.
