摘 要:利用化学法合成吗啡人工抗原,将此抗原免疫新西兰大白兔制备抗吗啡多克隆抗体.利用混合酸酐法将吗啡人工抗原分别与蛋白质载体BSA及OVA偶联,经蛋白质电泳以及紫外扫描检测,证明吗啡人工抗原合成成功.用间酶联免疫检测法检测,证明制备得到了高效价的吗啡多克隆抗体血清,且与吗啡人工抗原具有高特异性反应,可用于吗啡免疫学检测方法的研究.Artificial morphine antigen was chemically synthesized and was then used to immunize the New Zealand rabbits to prepare the polyclinal anti-morphine antibody. Afterwards, the morphine antigen is respectively coupled with two protein carriers ( BSA and OVA) by means of mixed acid anhydride method. It is proved by UV scan and protein electrophoresis that the artificial morphine antigen is successfully synthesized. Moreover, it is proved by ELISA results that the rabbit serum with high dilute degree is prepared, and that there is a high-specificity reaction between the polyclonal antibody and the antigen. Both the antigen and the antibody can be applied to the research on morphine immunoassay.
