摘  要:为了探讨肾脏近曲小管上皮细胞囊泡膜H+-ATP酶的细胞化学定位,我们在中性环境中,以Tricine-KOH为缓冲浪,P-NPP为底物,用饰作捕捉剂,分组进行了实验。结果:溶酶体内的酶活性可被NaF所抑制,溶酶体和内吞体股上的酶活性则不能被NaF抑制,但能被NEM所抑制。实验提示:溶酶体和内吞体股上的P-NPP酶,是对K+依赖的,对ouabain不敏感的,但可被NEM抑制。This work was attempted to study cytochemically the localization of H+-ATPase on the vacuole membranein the epithelial cells of renal proximal tubules. In the incubation medium, P-NPP was used as the substrate, cerium asthe capture agent, and tricine-KOH as buffer to make a neutral condition. As a result, it is found that the enzyme activity Inside the matrix of lysosome can be inhibited by NaF, but insensitive to the enzyme activity on the lysosome and endosome membrane, the enzyme activity can be inhibited by NEM. The finding suggested that the enzyme activity on themembrane of lysosome and endosome was ouabain-insensitive and NEM-inhibitory proton pump H+-ATPase.
