摘  要:目的探讨运动对内源性二甲基精氨酸二甲胺水解酶(DDAH)活性及表达的影响与骨质疏松关系。方法3月龄雌性SD大鼠40只,骨质疏松模型组切除双侧卵巢。6 w后,每组随机选7只大鼠处死,取胫骨上段骨组织,观察模型复制情况。模型复制成后,随机分为3组:对照组,骨质疏松组,骨质疏松+运动(游泳)组;正常饲养6 w。饲养12 w后处死;剪下股骨远端和胫骨近端关节,冰浴下高速匀浆机匀浆,得酶匀浆,酶匀浆中的DDAH与ADMA反应,在530 nm比色测定DDAH的酶活性,用SDS-聚丙烯烯胺凝胶电泳及Western blotting印迹分析法测定二甲基精氨酸二甲胺水解酶表达。结果对照组大鼠骨组织DDAH的酶活性为(1.619±0.233)mmol.L-1.g-1Pro.min-1。骨质疏松组大鼠骨组织DDAH的酶活性(1.204±0.330)mmol.L-1.g-1Pro.min-1。运动组(1.473±0.196)mmol.L-1.g-1Pro.min-1。对照组大鼠骨组织DDAH的酶活性高于骨质疏松组。各组DDAH酶表达无差异。结论骨质疏质时骨组织中DDAH的酶活性含量降低,运动可抑制酶活性降低。骨组织DDAH的酶活性与OP有密切的关系。Objective: research on the influence of spots on the activity of osteoporosis endogenous ADMA&DDAH hydrolyzedenzyme. Method: Forty female rats of three months' old were served as the subjects of the experiment, who were local anesthesia at the abdomen cavity. To the compared group, a bit of fat from some of rats was resected, while to the subjects both side of ovaries were resected in the course, of comparison. Six weeks later, death was put to seven of each group at random and a piece of the bone tissue fl'om the upside shank bone was picked. The situation of the model copied was observed. After copying accomplished, the model subjects were randomly divided into three groups: Comparison Group, Osteoporosis Group and Osteoporosis & Sport Group, From then on, they were fed normally for 6 weeks and then were killed 12 weeks later. The arthrosis of the far end of the thighbone and the near end of the shank bone were picked and were made into liquid form. The activity botween DDAH and ADMA was observed, and the DDAH enzyme activity would be illustrated by the amount of L- Citrulline produced in 530nm. Result: From our observation of the selected subjects of the different groups, we made the conclusion that the sport activity could keep the content of ADMA&DDAH hydrolyzed - enzyme from rising due to the fact that the enzyme activity of DDHA has a close relationship with OP.
