摘  要:目的观察痛必定注射液对坐骨神经结扎性慢性损伤模型(CCI)大鼠脊髓N-甲基-D-天冬氨酸受体1(NR1)的影响。方法雄性SD大鼠30只,随机分为正常组、CCI组和痛必定组。采用RT-PCR技术测定各组大鼠脊髓内NR1表达的变化。结果CCI组较正常组脊髓内NR1mRNA表达量明显升高(P<0.001);CCI+痛必定组NR1mRNA表达受到明显抑制(P<0.001)。结论痛必定注射液在脊髓水平的镇痛机理与其抑制NR1mRNA的表达有关。Objective To study the effect of Tongbiding injection on expression of N-methyl-D-aspartate (NMDA) receptor 1 mRNA in the spinal cord of chronic constriction injury (CCI) rats. Methods Thirty male Spragru-Dawley rats were randomly divided into control group (group A), CCI group (group B) and Tongbiding group (group C), ten rat for eah group. The changes in NR1 mRNA expression in the spinal cord was detected by reverse transcription- polymerase chain reaction (RT-PCR) techniques. Results The expression of NR1 mRNA in the spinal cord was minimal in control group, CCI induced significant increase in the expression of NR1 mRNA in the spinal cord and Tongbiding significantly inhibited the increase in NR1 mRNA expression. There was significant difference between CCI and Tongbiding group (P〈0.001). Conclusions Inhibition NR1 mRNA expression in the CCI rats spinal cord may contribute to analgesic mechanisms of Tongbiding in the spinal dorsal horn.
