摘 要:目的:了解广西金秀瑶族群体12个STR基因座的遗传多态性。方法:对175例瑶族男女抽取外周血,采用Chelex-100法提取DNA,荧光标记复合扩增基因座,ABIPrism(3100型遗传分析仪对扩增产物进行检测。结果:12个位点(D2S1338、D3S1358、D5S818、D7S820、D8S1179、D13S317、D16S539、D18S51、D19S433、D21S11、FGA、vWA)共检测出94种等位基因,基因频率分布在0.0029~0.3943;322种基因型;符合Hardy-Weinberg平衡定律;观察杂合度(HO)分布在0.7371~0.9029,个人识别力(DP)分布在0.8753~0.9601,累积个人识别力(TDP)为>0.999999999,非父排除率(EP)在0.6046~0.8519,累积非父排除率(CEP)为0.999999974,多态信息量(PIC)分布在0.6788~0.8428。结论:广西金秀瑶族12个STR基因座均属于高度多态性遗传标记。Objective: To study the STR genetic structure of JinxiuYao ethnic group in Guangxi. Methods: Distribution for 12 STR loci was determined in Jinxiu Yao ethnic group based on STR genescan marked by fluorescence. Result:94 alleles and 322 genotypes were observed in 175 unrelated Jinxiu Yao individuals, with the gene frequency being 0.002 9-0. 394 3, respectively. The genotypes of 12 STR loci were in accordance with the Hardy-Weinberg equilibrium ( P 〉0.05). The statistical analysis of 12 STR loci showed PIC(polymorphism information content) 0. 678 8-0. 842 8, H(heterozyaosity) 0. 737 1-0. 902 9, DP(discrimination power) 0. 875 3-0. 960 1, EP(Excluding probability of paternity) 0. 604 6-0. 851 9. According to the obtained data, the cumulative exclusion power and for these STR markers in Jinxiu population is 0. 999 999 974 and the cumulative discrimination power is 〉0. 999 999 999,respectively. Conclusion: The 12 STR loci of Jinxiu Yao ethnic group in Guangxi shows high genetic polymorphism. These results play an important role in Chinese population genetic study.
