摘  要:以萱草属的9个野生种和1个变种为材料,应用ISSR-PCR分子标记技术,对野生种间的亲缘关系和多态性进行了研究。从123条ISSR通用引物中筛选出了10条扩增产物清晰且多态性高的引物,对实验样本进行扩增,共产生了218条条带,多态率为100%,野生种间遗传多态性丰富。采用TFPGA数据分析软件,计算出Nei's相似系数和遗传距离,构建亲缘关系树状聚类图。野生种之间的遗传距离为0.0187~0.4941,种间遗传差异明显。其中,北黄花菜与黄花菜的遗传距离最小,矮萱草与黄花萱草之间的遗传距离最大。亲缘关系聚类图显示各种被聚为4组:北黄花菜、黄花某、萱草、小萱草、重瓣萱草为一组;西南萱草、折叶萱草和小黄花菜聚为另一组;黄花萱草和矮萱草各为一组。因此,矮萱草与其他种间亲缘关系最远;黄花萱草其次;黄花菜与北黄花菜亲缘关系最近;萱草、小萱草、重瓣萱草亲缘关系接近;西南萱草与折叶萱草、小黄花某亲缘关系接近。The ISSR-PCR marker was used to study relationships and genetic polymorphism for nine species and one variation of Hemerocallis .Ten primers were screened from 123 random primers. A total of 218 DNA fragments were amplified. They were polymorphic. The molecular evidence indicated that different species in the genus were high polymorphism. Nei'similarity coefficients and genetic distances was calculated by TFPGA. The dendrogram of nine species and one variation in the genus can be constructed by UPGMA. The results indicated that the nine species and one variation of the Hemerocallis can be divided into four groups. H. lilioasphode,H, citrina,H, fulva,H, dumortieri, H. fulva var. kwanso were in the first group; The second group was the cluster of H. forrestii, H. plicata and H. minor. The third group was H. flava, the fourth group was H. nana. The genetic distance among the different species ranged from 0. 0187 to 0. 4941. The closest relationship was H. lilioasphode and H. citrina. The next was H. forrestii with H. plicata and H. minor. H. fulva, H. durnortieri,H, fulva var. kwanso were coming after that. H. nana kept the further genetic distance with other daylily species.
