摘  要:目的:探讨莱菔子水溶性生物碱对自发性高血压大鼠(SHR)的降压作用以及对大鼠血清一氧化氮(NO)、超氧化物歧化酶(SOD)、丙二醛(MDA)及心肌一氧化氮合酶(NOS)活性的影响。方法:将40只SHR随机分为5组:空白对照组、莱菔子水溶性生物碱高、中、低剂量组、卡托普利组,每组8只。采用尾动脉测压法测量给药前及给药后2、4、6、8w大鼠收缩压,于给药8w后,检测大鼠血清NO含量,SOD活性,MDA含量以及心肌NOS活性。结果:给药8w后,莱菔子水溶性生物碱明显降低了SHR的血压,其中高、中剂量组的降压幅度与卡托普利组比较无显著性差异,同时莱菔子和卡托普利均能增加大鼠心肌NOS活性和血清NO含量,并且莱菔子水溶性生物碱能显著提高大鼠血清SOD活性,降低MDA含量,而卡托普利此作用不明显。结论:莱菔子降压作用可能与激活了NO-NOS系统有关,并可能通过抗氧化起到保护靶器官的作用。Objective. To explore the effects of the soluble Semen Raphani alkaloids on spontaneous hypertensive rats (SHR), their serum nitric oxide (NO)content, superoxide dismutase (SOD) activity, content of malondialdehyde (MDA) and cardiac nitric oxide synthase (NOS) activity. Methods: Forty SHRs were randomly divided into five groups: untreated control group, the high, middle, low dosage group of the Semen Raphani soluble alkaloids, and Captopril group, with eight rats in each group. SBP were measured respectively by tail-cuff technique before therapy and two weeks, four weeks, six weeks, eight weeks after therapy. The serum content of NO, activity of SOD, content of MDA and activity of NOS in cardiac tissue were measured after eight weeks therapy. Result: Eight weeks treatment later, the soluble alkaloids of Semen Raphani significantly declined the blood pressure of rats. Compared with that of Captopril, the hypotensive function of high and middle dosage groups had no significant difference. Meanwhile, both Semen Raphani and Captopril significantly enhanced serum NO content and cardiac NOS activity of SHRs. Besides, Semen Raphani decreased their serum MDA content and enhanced SOD activity greatly, while Captopril had no this effect. Conclusion. The hypotensive function of the Semen Raphani may be related to its NO-NOS system activation, and may protect target organ via antioxidation.
