黄芪失笑散对鸡胚绒毛尿囊膜促血管生成实验研究

摘　　要：目的:通过鸡胚绒毛尿囊膜(chrioallantoic membrane,CAM)模型以探讨黄芪失笑散的促血管生成的作用及机理,为进一步开发及临床应用提供实验依据。方法:取SD大鼠16只随机分成模型组、消心痛组、黄芪失笑散大剂量组,消心痛和黄芪失笑散大剂量组分别进行灌胃10天,末次灌胃后1.5h取血制成含药物的血清,黄芪失笑散小剂量组药物血清以黄芪失笑散大剂量和模型组等剂量配置而成。然后分别以贝复济组(碱性成纤维细胞生长因子bFGF)和模型组分别为阳性和阴性对照组作用CAM,通过进行一级及二级血管计数来评价黄芪失笑散的促血管生成活性并初步探讨其作用机理。结果:黄芪失笑散大剂量在CAM一级及二级血管数目上均与贝复济组间比较无显著性差异(P>0.05)。结论:黄芪失笑散具有显著促血管生成作用。Objective ： The experiment investigates the mechanism of Astragalus Shixiao decoction for promoting anglogenesis on chrioallantoic membrane （ CAM）, and provides an experimental basis for further developing and using it in clinic. Methods： Sixteen SD rats were randomly divided into four groups： model group, Isosorbide dinitrate group, large dosage Astragalus Shixiao decoction group, Isosorbide dinitrate adding large - dosage Astragalus Shixiao decoction group. The respective dose and drugs were given to the treated groups, the same dose of distilled water was given to model group. Blood serum of containing drugs were got after ten days and were put on the chick embryo that had been hatched nine days, while bFGF and distilled water as controlled. Chorioallantoic membrane was segregated from egg shell on the 12th day, and the number of first and second class of blood vessels were counted by picture analytical system. Results： The number of the first and second class of blood vessels increased similarly in large - dosage Astragalus Shixiao decoction group, compared with bFGF group（ P 〉 0.05）. Conclusion： Astragalus Shixiao decoction can promot Angiogenesis on CAM notably.
