摘 要:为了研究下丘脑EOP系统在猪应激反应过程中的作用,本试验随机选取12头二花脸生长猪,随机分为对照组和运输组,运输组进行2 h的运输,运输结束后15 min,运输组和对照组同时采样。以18S rRNA或GAPDH mRNA为内标,用相对定量RT-PCR方法检测下丘脑中前阿黑皮素原(Proopiomelanocortin,POMC)、激素前体转化酶2(Prohormone Convertase 2,PC2)、μ阿片肽受体(Mu Opioid Receptor,MOR)和糖皮质激素受体(Glucocorticoid Receptor,GR)mRNA的表达。结果表明:下丘脑POMC mRNA表达水平运输组猪极显著高于对照组;MOR mRNA的表达运输组显著高于对照组;PC2和GR的mRNA表达和对照组相比,差异不显著。提示二花脸生长猪下丘脑EOP系统参与了运输应激反应调节。In order to evaluate the role of hypothalamic EOP system in the stress response of pigs, 12 growing Erhualian (EHL) pigs were randomly divided into the control and transport groups. The transport group was subjected to 2 h road transportation, and 15 min after transportation, pigs from both groups were sampled. The expressions of mRNA for proopiomelanocortin (POMC), prohormone convertase 2 (PC2), mu opioid receptor (MOR) and glucocorticoid receptor (GR) in the hypothalamus were determined by semi-quantitative RT-PCR based on the internal standards of 18S rRNA or GAPDH mRNA. The results showed that the expression of POMC mRNA in the transport group was significantly higher than that in the control group, and the expression of MOR mRNA in the transport group was significantly higher than that in the control group. However, there was not significant difference in the expressions of GR mRNA and PC2 mRNA between the control and the transport groups. The above results indicated the participation of the pigs EOP system in the regulatory mechanism in response to transport stress.
