摘  要:目的:探讨加味大柴胡汤对阻塞性黄疸大鼠肝脏MRP2表达的影响及其对肝脏的保护机制。方法:将54只大鼠随机分三组,即:假手术+生理盐水灌胃组(SO+NS组)、胆总管结扎+生理盐水灌胃组(BDL+NS组)、胆总管结扎+加味大柴胡汤灌胃组(BDL+MMDB组)。分别在术后7、14、21d取肝组织标本,用PV-6001免疫组织化学法和半定量RT-PCR法检测肝脏MRP2蛋白和MRP2mRNA的表达。结果:各时相段中BDL+MMDB组的MRP2蛋白和MRP2mRNA的表达低于SO组,而明显高于BDL+NS组,差异均有统计学意义(P<0.01)。结论:加味大柴胡汤能上调MRP2的表达,减轻肝脏的损伤。Objective: Effects of modified major decoction of Bupleurum(MMDB) on MRP2 of liver in rat with obstructive jaundice are observed,which prove that MMDB can improve the impairment of liver in rat with obstructive jaundice.Method: 54 rats were divided into three time points at random,namely SO group,BDL+NS group,BDL+MMDB group.The rats were sacrificed at 7th,14th,21st day in order that liver tissue were taken out,subsequently.The level of MRP2 protein and MRP2 mRNA in the liver were tested with immunochemistry and reverse transcription PCR.Result: At different time point the level of MRP2 protein and MRP2 mRNA in the liver in BDL+MMDB group were lower than in SO group,but were obviously highltr than those in BDL+NS group,Comparison in statistics among BDL+MMDB group,SO group and BDL+NS group were highly significant(P〈0.01).Conclusion: The level of MRP2 protein and MRP2 mRNA in the liver in rat with obstructive jaundice could be up-regulation by modified major decoction of Bupleurum,and it could relieve the impairment of liver.
