摘 要:目的观察髓核组织Fas和Bcl-2蛋白的表达,探讨其在腰椎间盘退变过程可能发挥的作用。方法采用原位DNA片断末端标记(TUNEL)和免疫组织化学方法,检测20例退变腰椎间盘(退变组)及6例正常腰椎间盘(对照组)髓核组织的细胞凋亡状态及凋亡相关基因Fas和Bcl-2的表达。结果退变组及对照组髓核组织细胞凋亡指数(AI)分别为(50.23±13.36)和(28.32±6.67)(P<0.05),Fas蛋白阳性细胞百分比(74.26±10.15)%和(41.72±4.62)%(P<0.01),Bcl-2蛋白阳性细胞百分比(55.37±13.65)%和(27.17±5.17)(P<0.01)。凋亡细胞越多,Fas和Bcl-2的表达越强。结论Fas和Bcl-2蛋白均参与腰椎间盘组织中细胞凋亡的调节,可能在腰椎间盘退变中发挥重要作用。Objective To investigate the relationship between expression of Fas and Bcl-2 protein in apoptotic status and lumbar intervertebral disc degeneration. Methods The apoptotic status and the expression of Fas and Bcl-2 in 20 cases of degenerative lumbar intervertebal disc tissues and 6 of normal lumar intervertebal disc tissues was detected by using TdT-mediated dUTP-biotin nick end labeling (TUNEL) and immunohistochemistry methods. was (50.23±13.36),(28.32±6.67)(P〈0. 05). The expressions rates of Bcl-2 protein in two groups The stronger Fas and Bcl-2 protein expressed, the Conclusion Both Fas and Bcl-2 protein probably take part in the apoptosis of nucleus pulposus and the progression of lumbar intervertebal disc degeneration.
