摘  要:目的研究志贺菌外排泵cmr基因的耐药作用。方法以志贺菌临床多耐药株H24基因组为模板,扩增出含cmr基因的1620 bp DNA片段,将所得片段与pMD18-T载体连接,转化到大肠埃希菌DH5α中,酶切鉴定阳性克隆,对其进行序列测定并用琼脂平皿二倍稀释法对重组菌株进行药敏测试和泵抑制剂实验。结果含有志贺菌cmr基因的大肠埃希菌较不含有该大肠埃希菌对红霉素和四环素的耐药性提高1倍,并且这种耐药性提高可被质子泵抑制剂氰氯苯腙(CCCP)所抑制。结论cmr基因与志贺菌耐药株H24对红霉素和四环素的耐药性相关。Objective To study the function of active efflux pump gene cmr in clinical isolates of Shigella. Methods The active efflux pump gene cmr of Shigella sonnei H24 was amplified by PCR and ligated with pMD18 - T to construct the recombinant plasmid pMD-cmr, and the pMD-cmr was transformed into E. coli DH5α. The positive recombinant plasmid was identified by restriction enzyme digestion and the gene was sequenced and analyzed. Drug susceptibility of the recombinant strain was tested by the two fold agar dilution technique; The MICs of the seven antibiotics were also determined by adding CCCP. Results The E. coli strain with cmr gene was more resistant to tetracycline and erythromycin than the strain without it, and the resistances could be inhibited by proton pump inhibitor CCCP. Conclusion The gene cmr is related with the resistance of tetracycline and erythromycin in the drug resistant - strain H24.
