摘 要:目的 证实胃饲乙醇预处理对重度肝脏缺血再灌注(ischemia reperfusion,IR)损伤耐受性的增强效应。方法健康成年雄性Wistar大鼠120只,分为正常组、生理盐水预处理组、胃饲乙醇预处理组(采用40%乙醇5g/kg胃饲预处理大鼠)。采用门静脉转流的肝脏缺血模型,缺血时间90min,于再灌注不同时相点观察动物肝功能、肝组织还原型谷胱甘肽含量、细胞凋亡及肝组织病理。结果胃饲乙醇预处理组从形态结构到肝功能,均比对照组有明显改善,可有效减少IR后肝细胞的坏死和凋亡,更大程度地保留肝组织还原型谷胱甘肽含量。结论胃饲乙醇预处理可有效增强肝脏对重度缺血再灌注损伤的耐受性,有望成为一种新的预处理措施。Objective To investigate the reinforcement effect of ethanol preconditioning on the tolerance of rat liver to the ischemia-reperfusion (IR) injuries. Methods A total of 120 Wistar rats were randomly divided into Group A ( normal control group, n = 8 ) , Group B ( rats pretreated with 0.9% normal saline at 5 g/kg, n = 56), and Group C (rats pretreated with 40% ethanol at 5 g/kg, n =56). Twenty-four hours after pretreatment, the hepatic blood inflow of rats in Group B and C was blocked for 90 minutes, and then the liver function, reduced glutathione hormone (GSH) content, hepatocyte apoptosis and pathologic changes of the liver were detected after reperfusion for 0, 1, 3, 6, 12, 24, 72 hours. Results After 90 minutes IR injuries of liver, better liver structure, less necrotic hepatocytes and more reservation of GSH were found in Group B and C than Group A. The contents of ALT and AST in Group B and C were higher than those in Group A, with statistical significance ( P 〈 0.01 ). Conclusions Ethanol preconditioning can enhance the tolerance of rat liver to the IR injuries, thus it may become a novel management to fight against the IR injuries of liver.
