白芍总苷对兔佐剂性关节炎滑膜细胞增殖的影响

摘　　要：目的观察不同浓度的甲氨蝶呤(MTX)和白芍总苷(TGP)在体外实验中对类风湿关节炎滑膜细胞增殖的影响。方法利用弗氏完全佐剂制备兔佐剂性关节炎模型。利用MTT比色法测定不同浓度的甲氨蝶呤和白芍总苷单独或联合作用于滑膜细胞,对滑膜细胞增殖的影响。结果白芍总苷在0.8-25.6 mg/L浓度范围内抑制兔佐剂性关节炎滑膜细胞的增殖。随白芍总苷浓度的增高,抑制作用增强。各用药组与对照组相比,对滑膜细胞的增殖均具有显著的抑制作用。TGP组与MTX小剂量组或MTX大剂量组相比,抑制率均相近。结论白芍总苷在0.8-25.6 mg/L浓度范围内对兔佐剂性关节炎滑膜细胞的增殖具有抑制作用,并且呈浓度依赖性。在体外实验中,甲氨蝶呤和白芍总苷单独或联合作用均可显著抑制兔佐剂性关节炎滑膜细胞的增殖,这可能是甲氨蝶呤和白芍总苷发挥对类风湿关节炎治疗作用的重要机制之一。Objective： This study is aimed to compare the inhibitory effects of total glucosides of paeony on fibroblast - like synoviocytes respectively, or combined with methotrexate. Method： Synovium was obtained from rabbit adjuvant arthritis models which were induced by Freund＇s complete adjuvant. Synoviocytes were incubated with different concentration of methotrexate and total glucosides of paeony respectively or combined. Inhibitory effects were determined by MTT colorimetric assay. Results ： Total glucosides of paeony in concentration of 0. 8 - 25.6 mg/L demonstrated marked dose - dependent inhibitory effect on fibroblast - like synoviocytes proliferation. Both total glucosides of paeony and methotrexate presented remarkerable inhibitory effects on synoviocytes proliferation. There are all significant by statistics in inhibitory rate between MTX small dose group and MTX large dose group with total glucosides of paeony . Conclusion- Total glucosides of paeony （ 0. 8 - 25.6 mg/L） has dose - dependent inhibitory effect on proliferation of fibroblast - like synoviocytes. The results of this study suggest that in vitro both total glucosides of paeony and methotrexate have remarkably suppressive effects on proliferation of fibroblast -like synoviocytes, which play a significant role in the pathogenesis of rheumatoid arthritis. As many studies have proved , total glucosides of paeony and methotrexate can potentially prevent or limit the progression of rheumatoid arthritis.
