活血降糖胶囊对T2DM大鼠主动脉粥样硬化血管壁核因子-κB表达的影响

摘　　要：目的观察活血降糖胶囊对2型糖尿病(T2DM)大鼠并发主动脉粥样硬化血管壁核因子-κB(NF-κB)表达水平的调控作用。方法选1月龄SD大鼠45只,随机取8只为空白组(灌服等量蒸馏水),其余为实验组,成模大鼠随机分为中药组[灌服活血降糖胶囊悬浮液1.44g/(kg.d)和模型组(灌服等量蒸馏水)。每天灌胃1次,连续治疗30d后,取主动脉做常规石蜡切片,免疫组化观察其核因子-κB(NF-κB)表达的程度。结果模型组主动脉内膜出现典型粥样硬化病变,活血降糖胶囊中药组动脉膜受损程度有所减轻。模型组主动脉血管壁NF-κB表达明显增加,活血降糖胶囊中药组均可降低其主动脉NF-κB表达(P<0.05)。结论活血降糖胶囊可降低动脉粥样硬化大鼠主动脉壁NF-κB表达的作用,发挥对抗血管壁的炎性损伤作用。Objective To observe the therapeutic effect of Huoxuejiangtang capsule on expression of nuclear factor kappaB（NF- κB） in vessel wall of Sprague Dawley rats with atheroeclerosis. Methods Forty - five one month old Sprague Dawley rats were randomly divided into two groups： 8 of them were as normal control group, fed with normal diet, and the others were established into type two diabetes ＆ atherosclerosis （AS） model successfully with a low- dose streptozotocin （STZ） and animal fed with high sugar and cholesterol for six weeks . The latter was randomly subdivided into HXJT group , and Model group . They were treated with saline , different does of HXJT [ 1.44g/（kg·d） ], and the other two with equal quantity distilled water： by gavage once a day for a month, then the morphological changes of aortic intima were observed by light microscope. Results The morphological observation showed that typical atherosclerotic lesions of aortic intima were appeared in the model group, while the degree of atherosclerotic lesions was obvious lighter in the groups treated with HXJT. Expression of NF - κB was higher in model group than that in the normal control group , which was lowered in two HCJT groups （P〈0.05）. Conclusion HXJT could reduce expression of NF - κB and protect aortic intima and interfere the occurrence of atherosclerotic lesion.
