摘 要:目的:探讨温阳化饮方对支气管哮喘寒饮蕴肺证大鼠模型的干预机制。方法:将60只大鼠随机分为正常对照组(A组)、模型1d组(B组)、模型3d组(C组)、模型7d组(D组)、沙丁胺醇组(E组)、温阳化饮方组(F组),每组10只。采用动物造模、随机对照的方法,观察大鼠一般体征、体重、肛温、血气分析、肺组织β-ARmRNA表达的变化。结果:B、C、D、E、F组大鼠体重、肛温均低于A组(P<0.05),F组的体重高于D组、E组(P<0.05),F组的肛温均高于B、C、D、E组(P<0.05)。在血气分析方面,D组与A组比较,氧分压(PaO2)、血氧饱和度(SaO2)、血氧含量(tO2)降低而二氧化碳分压(PaCO2)升高(P<0.05或P<0.01);F组、E组与A组比较,PaCO2升高(P<0.05);E组与D组比较,tO2升高(P<0.05);F组与D组比较,SaO2、tO2升高(P<0.05);D组与A组相比,血液pH值、实际碳酸盐(AB)、标准碳酸盐(SB)以及标准碱剩余(SBE)均明显降低(P<0.05或P<0.01);F组、E组与B、C、D组比较,pH、SB、AB、SBE均升高(P<0.01或P<0.05)。在β-ARmRNA表达方面,B、C、D、E、F组低于A组(P<0.05或P<0.01),F组高于D、E组(P<0.01)。结论:温阳化饮方对支气管哮喘寒饮蕴肺证大鼠模型的干预机制,可能在于其能提高体温,增加体重,上调β-ARmRNA表达,从而促进β-AR功能重建,上调β-AR,改善肺通气,减轻组织缺氧和二氧化碳潴留,保护肺组织而减少损伤。Objective:To probe the mechanism of Wenyang Huayin (warm yang to dissolve damp) Decoc- tion on rats with bronchial asthma in the syndrome of cold fluid retention in lung. Methods:Sixty rats were randomized into normal control group (A),model 1 d group (B),model 3 d group (C),model 7 d group (D) ,albuterol group (E) ,and Wenyang Huayin Decoction group (F) ,with ten in each. Models were made for observing the changes of general signs, body weight, rectal temperature, blood gas analysis, and expression of β-ARmRNA of lung tissues. Results:Compared with Group A, the body weight and rectal temperature in group B,C,D,E and F were lower (P〈0.05). The body weight in Group F was higher than that of group D and E (P〈0.05) and the rectal temperature higher than that of Group B,C,D and E (P〈0.05). In blood gas analysis, compared with Group A, PaO2, SaO2 and tO2 in Group D decreased but PaCO2 increased (P〈0.05 or P〈0.01) ;PaCO2 in Group F and E increased (P〈0.05). tO2 in Group E increased compared with Group D ( P〈0.05). SaO2 and tO2 in Group F increased compared with Group D ( P〈0.05). Compared with Group A, PH, AB, SB and SBE in group D decreased obviously ( P〈0.05 or P〈0.01). PH,SB,AB and SBE in Group F and E increased compared with Group B,C and D (P〈0.01 or P〈0.05). In expression of β-ARmRNA,it was lower in Group B,C,D,E and F compared with Group A (P〈0.05 or P〈0.01);it was higher in Group F than that of Group D and E (P〈0.01). Conclusions:The mechanism of Wenyang Huayin Decoction on rats with bronchial asthma in the syndrome of cold fluid retention in lung may be in increasing the body temperature and weight and expression of β-ARmRNA,therefore β-AR function is rebuilt,ventilation of lung promoted,hypoxia of tissues and retention of carbon dioxide relieved,lung tissues protected and injuries reduced.
