草药肾病患儿肾组织内皮素-1及其受体基因的表达

摘  要:目的探讨中草药肾病患儿肾组织内皮素-1(ET-1)升高的效应部位以及效应的ET受体(ETR)亚型。方法采用斑点杂交、原位杂交和放射免疫技术对中草药肾病ET-1受体的基因表达和ET-1含量进行了研究。结果中草药肾病ET-1、ETA、ETB、mRNA均明显升高;ET-1mRNA主要分布肾皮质小血管内皮细胞、髓质肾小管和集合管,ETA受体mRNA则分布于上述小血管的平滑肌细胞;ETB受体主要分布髓质肾小管、集合管;中草药肾病血清和尿ET-1含量分别为92.56±32.11 pg/m l和23.78±8.33 ng/mmol均高于对照组65.12±21.36 pg/m l和11.56±6.11 ng/mmol(P<0.05)。结论增强表达的ET-1结合可能导致肾皮质缺血和水钠代谢异常,是中草药肾病后肾功能障碍的重要因素。Objective To study endothelin - 1 ( ET - 1 ) and its receptor subunits ( ETA, ETB) mRNA expressions in Chinese herbs nephropa- thy. Methods Northern blot hybridization, in situ hybridization and radioimmunoassay were used in this study. Results ET - 1, ETA and ETB mRNA expressions were all evidently increased. ET - 1 mRNA distributed primarily on glomerular endothelial cells and tubular epithelial cells in cortex; ETA mRNA predominantly presented on smooth muscle cells of small arterioles in renal cortex. However, ETB mRNA expression mainly localized on renal medullary tubular epithelial and collecting duct cells. Serum and urinary ET - 1 content (92.56 ± 32.11 pg/ml and 23.78 ± 8. 33 ng/mmol respectively) of Chinese herbs nephropathy group were higher than those (65.12 ± 21.36 pg/ml and 11.56 ± 6.11 ng/mmol respectively) in control group ( P 〈 0.05 ). Conclnsian Increased ET - 1 and its receptors in Chinese herbs nephropathy may play an important role in the pathogenesis and development of renal function impairment.
