摘 要:Ryanodine受体(RyR)是肌质网快速释放Ca2+的通道,在细胞钙信号转导及相关生理过程如肌细胞的兴奋-收缩偶联过程中扮演重要角色。SDS-PAGE和免疫印迹实验结果显示H2O2能够氧化SR膜上的RyR1,使其560-kDa单体和410-kDa内源性酶解片段的强度降低直至消失,并产生三个高分子量复合物。加入还原剂二巯基乙醇,能够抵消这种氧化作用。Ryanodine receptor ( RyR ) is the fast Ca^2+ release channel of the sarcoplasmic reticulum and plays a vital role in Ca^2+ signaling and excitation-contraction coupling in muscle cells. Upon treatment ofsarcoplasmic reticulum with H2O2, SDS-PAGE showed that the intensity of the 565-kDa RyR1 peptided and its 410-kDa endogenous enzymatic fragment (fRyR) was dramatically reduced, and in parallel, three high-molecular-weight bands were observed. Western blotting analysis revealed that these high-molecular-weight bands were cross-linked RyR subunits. The chemical changes of RyR1 were reversible upon reduction by reducing agent 2-mercaptoethanol.
