摘  要:目的探讨川芎嗪和地塞米松对活动期类风湿关节炎(RA)患者外周血单个核细胞(PBMC)B7-1和B7-2mRNA表达的影响,为临床使用川芎嗪治疗RA提供理论依据。方法取活动期RA患者PBMC培养,分别以川芎嗪、地塞米松和川芎嗪+地塞米松进行处理,以PBS对照,采用半定量逆转录PCR的方法,检测PBMCB7-1和B7-2mRNA表达。结果与对照组相比,川芎嗪组和地塞米松组均能明显抑制B7-2mRNA表达(P<0.05),而川芎嗪+地塞米松组对B7-2mRNA表达的抑制更加明显(P<0.01)。各组对B7-1mRNA表达均无显著影响(P>0.05)。结论川芎嗪能下调活动期患者PBMCB7-2mRNA表达,合并使用地塞米松能更好地抑制B7-2mRNA表达。Objective To explore the effect of ligustrazin and dexamethasone on expressions of B7-1 (CD80) and B7-2 (CD86) mRNA in peripheral blood mononuclear cells (PBMCs) from active stage rheumatoid arthritis( RA)so as to obtain some principle evidences of RA treated with ligustrazin. Methods PBMCs from active stage RA were cultivated and treated with ligustrazin, dexamethasone and ligu- strazin plus dexamethasone to compare those treated with PBS. B7-1 and B7-2 mRNA expressions were measured by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). Results Ligustrazin and dexamethasone respectively inhibited expressions of B7-2 mRNA (P 〈0. 05) significantly, especially in ligustrazin plus dexamethasone group (P 〈0.01 ). But BT-1 mRNA expression of all groups had no obvious change ( P 〉 0. 05 ). Conclusions Ligustrazin could significantly down-regulate expression of B7-2 mRNA in active stage RA and has synergetie effect with dexamethasone.
