摘  要:目的探讨β1转化生长因子(TGF-β1)对瘢痕疙瘩成纤维细胞分泌基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶-2(MMP-2)和金属蛋白酶组织抑制剂-1(TIMP-1)表达的调节作用,及TGF-β1与瘢痕的关系。方法培养的瘢痕疙瘩成纤维细胞经TGF-β1处理,采用酶联免疫法(ELISA)测定瘢痕疙瘩成纤维细胞上清液中MMP-1、MMP-2和TIMP-1的水平。结果瘢痕疙瘩中成纤维细胞MMP-1、MMP-2和TIMP-1的生成量显著高于正常皮肤(P〈0.01);加TGF-β1处理后,瘢痕疙瘩中成纤维细胞MMP-1的分泌量显著降低(P〈0.01),MMP-2的分泌量显著升高(P〈0.01),而TIMP-1的分泌量无显著改变(P〉0.05)。结论培养的瘢痕疙瘩成纤维细胞显示MMP-1、MMP-2和TIMP-1增加,TGF-β1在生成调节过程中起重要作用。Objective To investigate the regulatory effects of transforming growth factor-β1 on the secretion of matrix metalloproteinase-1 (MMP-1), matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in human keloid-derived fibroblasts. Methods The cultured keloid fibroblasts were treated with TGF-β1. Enzyme-linked immunosorbent assay (ELISA) method were performed to measure the production of MMP-1, MMP-2 and TIMP-1 in human keloid fibroblasts supernatant. Results The production of MMP-1, MMP-2 and TIMP-1 in human keloid fibroblasts were significantly higher than those in normal dermal fibroblasts (P〈0.01); after TGF-β1 was added to the cultures, the production of MMP-1 was decreased significantly (P〈0.01), the production of MMP-2 was increased significantly (P〈0.01), and production of TIMP-1 did not change significantly (P〉0.05). Conclusions Cultured keloid fibroblasts show increased production of MMPs, and TGF-β1 plays a role in this regulation of production.
