缬沙坦对脓毒症大鼠的早期干预作用

摘　　要：目的探讨缬沙坦对脓毒症大鼠的早期干预作用。方法大鼠72只,随机分为4组:正常组、模型组、缬沙坦干预组、生理盐水对照组。每组分别于腹腔注射后2h、4h、8h三个时间点(T_2、T_4、T_8)进行相关检测。正常组仅予生理盐水3 mg/kg腹腔注射,模型组予腹腔注射脂多糖(LPS,3 mg/kg),缬沙坦干预组在腹腔注射LPS后10 min予缬沙坦(5 mg/kg)灌胃,生理盐水对照组在腹腔注射LPS后10 min予等量生理盐水灌胃。分别于腹腔注射LPS或生理盐水后相应时间点处死动物,取心脏血,采用ELISA法检测各组血清中肿瘤坏死因子(TNF)-α、血管内皮生长因子(VEGF)、可溶性L-选择素(sL-seleetin)的浓度。结果大鼠腹腔注射LPS后,血清TNF-α、VEGF、sL-selectin的浓度均有明显升高。缬沙坦干预组与模型组各时间点相比,上述增高的细胞因子浓度显著降低(P<0.05)。结论缬沙坦能显著降低脓毒症大鼠增高的血清TNF-α、VEGF、sL-seleetin的浓度水平,通过减少炎症相关介质的释放、抑制白细胞活化、降低毛细血管通透性等机制减轻机体的炎症反应。Objective To investigate the effect of early intervention of valsartan on septic rats. Methods Seventy-two rats were randomly divided into four groups： normal control group, model group, valsartan group, normal saline group. The rats of normal control group were given normal saline （3 mg/kg） through peritoneal injection. The rats of model group were given LPS （3 mg/kg） through the same way. The rats of valsartan group were lavaged with valsartan （5 mg/kg） ten minites after LPS peritoneal injection while the rats of normal saline group were lavaged with physiologic saline. The rats were sacrificed at corresponding time after LPS or normal saline were injected peritoneally, then blood samples were collected from heart quickly. Serum TNF-α,VEGF and sL-selectin were detected by ELISA. Results For the rats given LPS through peritoneal injection, serum TNF-α, VEGF and sL-selectin increased significantly. However, serum TNF-α, VEGF and sL-selectin decreased obviously in valsartan group as compared with model group （P 〈 0.05 ）. Conclusion Valsartan can decrease the levels of TNF-α,VEGF and sL-selectin obviously in septic rats. It reduced systemic inflammatory response by lowering inflammatory mediator levels and inhibiting the leukocyte activation to decrease the capillary permeability.
