壮肝逐瘀煎对肝纤维化大鼠肝Smads表达的影响

摘　　要：目的:研究壮肝逐瘀煎对肝纤维化(HF)大鼠Smad3,Smad4,Smad7表达的影响,探讨其抗HF的作用机制。方法:采用CCl4复合因素法建立Wistar大鼠肝纤维化模型。对肝纤维化大鼠予壮肝逐瘀煎灌胃治疗,并予秋水仙碱及大黄虫丸为对照。12周后获取肝组织,苏木精-伊红染色观察肝纤维化的程度;免疫组织化学染色、图像分析方法对Smad3,Smad4,Smad7的组织分布进行半定量分析。结果:与病理模型组大鼠相比,壮肝逐瘀煎治疗组肝小叶结构趋于正常,纤维间隔明显变薄,肝组织Smad3、Smad4表达显著减少(均P<0.01),Smad7表达显著增加(P<0.01)。结论:壮肝逐瘀煎能够显著改善肝纤维化大鼠肝组织的病理变化,具有明显的抗肝纤维化作用,其作用机制可能与壮肝逐瘀煎调控Smad3,Smad4,Smad7的表达有关。Objective： To study the effects of Zhuanggan Zhuyu Jian on expression of Smad3, Smad4 and Smad7 in rats with hepatic fibrosis and explore the action mechanism of anti - hepatic fibrosis. Methods： The Wistar models of rats with hepatic fibrosis were made by CCl4 compound factors. The treatment with Zhuanggan Zhuyu Jian through administration by gavage was compared with the control group with Colchicines and another control group with Dahuang Zhechong Wan. Liver tissue was sampled at week12, to observe the level of hepatic fibrosis by using HE staining and tissue distribution of Smad3, Smad4 and Smad7 was semi - quantitatively analyzed by using immuno - histochemical staining and image analysis. Results： Compared with the pathological model group, the structure of hepatic lobules tended to be normal and the fibrous septum was significantly thinner; the expression of Smad3 and Smas4 in liver tissue was markedly lower （P 〈 0.01 ） and expression of Smad7 was pronotmcedly higher（P 〈 0.01） in the treatment group with Zhuanggan Zhuyu Jian. Conclusions： Zhuanggan Zhuyu Jian can improve the pathological change in liver tissue of rats with hepatic fibrosis and has significant action of anti - hepatic fibrosis.The action mechanism might be associated with its regulatory effects on the expression of Smad3, Smad4 and Smad7.
