摘  要:目的探究京尼平(C11H14O5)交联明胶的特性及其复合颗粒骨修复骨缺损的效果。方法从交联度、细胞毒性、溶胀度、体外降解率等方面,观察交联时间和京尼平浓度对京尼平交联明胶特性的影响。以兔桡骨缺损为模型,分为空白组、颗粒骨组、黏合颗粒骨组,通过X线、组织学切片观察各组骨缺损修复情况。结果随着交联时间和京尼平浓度增加,交联度增加,溶胀度和降解率降低;各组材料细胞毒性均很低;京尼平交联明胶可有效黏合颗粒骨并修复兔桡骨缺损。结论京尼平交联明胶材料细胞毒性很低,其特性可随交联时间和京尼平浓度调节;适宜比例的明胶、京尼平可黏合颗粒骨并修复兔桡骨缺损。Objective To investigate the characteristics of genipin-crosslinked gelatin and its effect in the treatment of bone defect with morselized bone. Methods The effects of crosslinking time and genipin concentration on the characteristics of genipin-crosslinked gelatin were observed in view of crosslinking index, cytotoxicity, swelling ratio and in vitro degradation rate. The rabbit radius defect model was established and 3 groups were involved, Model group, Morselized bone group, Pasted morselized bone group. Bone defect restoration in each group was observed by X-ray radiography and histological sectioning. Results As the crosslinking time and genipin concentration increased, the crosslinking index was increased, and the swelling ratio and degradation rate were decreased, with low cytotoxicity of either group. Genipin crosslinking gelatin could paste morselized bone effectively and repair rabbit radius defect successfully. Conclusions The cytotoxicity of genipin crosslinked gelatin was low and its characteristics can be modulated by crosslinking time and genipin concentration. Gelatin and genipin at an appropriate proportion can paste morselized bone to repair rabbit radius defect.
