摘 要:目的本实验利用微创方法建立大鼠离体肺灌注模型。方法将一只受体大鼠股动脉、颈静脉插管,引流出静脉血灌注低温保存后的供体肺,氧合后的血液回输入受体大鼠体内。每只受体大鼠建立控制性交叉循环后,灌注一只供体肺1 h,取股静脉流出血样测定pH、PO2、PCO2和Hb的变化情况。结果各组间pH、PO2、PCO2和Hb经统计学分析无明显差异(P>0.05)。结论控制性交叉循环下受体大鼠血流动力学稳定,能够较长时间地提供符合生理状态的静脉化全血。可用于供体肺保存及缺血再灌注损伤的研究。OBJECTIVE To establish and modify of the isolated lung transplantation and reperfusion model in rats with a relatively less - invasive way. METHODS Angiocatheters were inserted into femoral artery and jugular vein of the recipient rat to establish the controlled cross - circulatory. The venous cannula was connected to the pulmonary artery of the ex vivo lung block after preservation and cold storage to provide inflow,and Pulmonary effluent blood from the lung block was collected and returned to the support rat via the femoral artery for detoxification and deoxygenation. Each support rat was used to perfuse one lung block for at least one hour. The inflow sample to each lung block was analyzed for pH, pO2, pCO2 , and Hb to follow alterations in support rat physiology. RESULTS There were no statistical differences in pH, PvO2, PvCO2 or Hb ( P 〉 0.05). CONCLUSION The support rat can deliver constant systemic venous blood at stable physiologic parameters to the ex vivo lung block and this animal model can be used in research of lung transplantation and ischemicreperfusion injury.
