摘 要:目的探讨附子生物碱薄层层析色谱(TLC)条件的影响因素,完善附子生物碱TLC的检查方法。方法采用单因素试验方法,对影响附子生物碱TLC的因素进行考察,筛选出最佳TLC条件。结果以正己烷-乙酸乙酯-95%乙醇(12.8∶7.2∶2)为展开剂,用浓氨水和展开剂同时饱和40min,展开,取出晾干,以10%浓硫酸-乙醇为显色剂,70℃下加热30min,可获得分离清晰的6个有色斑点。结论浓氨水和展开剂同时饱和40min,展开,显色,可获得较好的分离效果。Objective To improve and optimize the method of thin-layer chromatography (TLC) for the alkaloid determination in radix aconite lateralis praeparata. Methods Several influential factors on TLC, such as the developers, time of saturation and concentration of strong aqua ammoniac, were investigated by using the single factor experimentation. Results N-hexane : acetoacetate : 95% alcohol(12.8:7. 2: 2. 2)were used as developing agent together with strong aqua ammoniae, in which the thin layer plate was saturated for 40 min. After drying, the plate was sprayed with concentrated sulfuric acid: alcohol(10% ). Six distinctly separated spots were obtained after the plate was heated in the oven for 30 min at 70℃, Conclusion The optimal chromatographic conditions are obtained. This method may be used for the alkaloid determination in radix aconite lateralis praeparata.
