摘  要:目的进行福尔马林诱导的大鼠内脏痛行为及N型钙通道阻断剂SNX-111镇痛效果观察和评价。方法采用福尔马林直肠黏膜下注射复制的内脏痛模型,鞘内注射SNX-111(0.1μg/kg;0.5μg/kg;1μg/kg)观察镇痛效果。致痛后以15min为一个时间段,共记录2h,分别计算疼痛分数。结果各组大鼠在注射福尔马林后30min均达到疼痛评分的最大值[(39.88±5.32)分-(89.84±8.61)分],从注射福尔马林后45-120min内,疼痛评分逐渐降低[(2.96±0.91)分-(38.50±5.16)分]。前60min内行为表现主要以伸展身体、腹部收缩、全身收缩反应为主;而在后60min内主要以腹部舔食和轻咬反应为主。SNX-111能明显减少疼痛评分[(4.48±0.45)分-(46.83±5.28)分],抑制内脏疼痛反应,SNX-111剂量越大,镇痛效果越显著(P〈0.01或P〈0.05)。结论福尔马林具有较强的内脏致痛效应,疼痛分数能反映内脏疼痛程度,可作为量化指标;SNX-111对内脏痛具有明显的、剂量依赖性的镇痛效果。Objective To observe and assess visceralgia behavior of the rat induced by formalin and anal-gesic effect of N-type calcium channel blocker SNX-111. Methods A model of visceralgia induced by formalin injection in the submucosa of rectum was used, SNX-111 (0. 1μg/kg;0.5μg/kg; 1μg/kg) was injected intrathe-cally to observe analgesic effect of it, A numerical score for pain behaviors was recorded every 15 minutes for 2 hours, Results The scores were at the highest level in 30-min period (39.88±5.32 - 89.84±8.61 ) and re- duced in late phases(2.96±0.91-38.50±5.16). Body stretching, contraction of the flanks and whole body contraction were predominant in the former 60 minutes, while abdominal licking and nibbling were predominant in the late 60 minutes. SNX-111 could significantly reduce pain scores (4.48±0.45 - 46.83±5.28 ) and alleviate pain reaction. The larger dose of SNX-111 used, the better analgesic effect had( P 〈 0.01, P 〈0.05 ), Conclusion Formalin has a significant algesie effect, and pain scores can reflect the degree of visceralgia and be a quan-tifiable index, SNX-111 has a significant and dose-dependant analgesic effect on visceralgia.
