甲醛固定组织中利多卡因检验

摘　　要：目的建立甲醛固定的生物组织中利多卡因含量的检测方法。方法采用甲醛防腐处理后脏器组织匀将去蛋白,有机溶剂分离提取,固相萃取净化后高效液相色谱法(HPLC)检测,并与新鲜组织中含量行对照研究。结果采用所建方法从实验动物新鲜组织和甲醛固定后组织及甲醛固定的目标人体脏器中全部检出利多卡因;按利多卡因100 mg.kg-1经腹腔注射给豚鼠,其检出量为x—±sD:心血-19.09±3.95μg.ml-1,新鲜脏器组织-2.20±0.58-3.82±0.80μg.g-1,甲醛固定3周脏器组织-1.91±0.60-3.30±0.77μg.g-1甲醛固定5周脏器组织-1.80±0.57-3.22±0.84μg.g-1;甲醛固定3-5 w,使其利多卡因检出量下降13.2%-22.9%。结论甲醛固定生物组织可以进行利多卡因含量测定,其检出量与给药量存在具有统计学意义的量变关系,能够作为相关案例的鉴定依据。Objective To establish the method of leidocaine detection in formalin fixed tissue for applying to the forensic toxicological analysis.Methods the tissue protein in the formalin fixed tissue was removed, and leidocaine was isolated with organic solvent, purified with solid phase extraction, and analyzed with HPLC,the fresh tissue was treated as control. Results leidocaine could be detected in the fresh tissue of animal and formalin fixed specimen of human. The guinea pig was administrated with 100 mg·kg^- 1 leidecaine, the leidocaine level was 19.09 ± 3.95 μg· ml^- 1 in blood, 2.20 ± 0.58 - 3.82 ± 0.80 μg· g^- 1 in fresh tissue, 1.91 ± 0.60 - 3.30 ± 0.77 μg· g^- 1 in formalin fixed tissue （3w）, 1.80 ± 0.57 - 3.22 ± 0.84 μg· g^- 1 in formalin fixed tissue （5w）. The leidocaine level in formalin fixed tissue （3 - 5w） decreased about 13.2% - 22.9%. Conclusion Leidocaine can be detected from the formalin fixed tissue,the level of Leidocaine was associated with the administrated dosage, it will be an important evidence of some case.
