摘  要:目的通过对流式细胞术测定HLA-B27实验中荧光强度随时间而变化的研究,寻找可用判断HLA-B27阴阳性的较合理方法。方法以HLA-B27阳性/HLA-B7阴性细胞占淋巴细胞90%以上,同时其MFI大于8作为阳性标准。选择32例阳性标本上机测定获得MFI值,此后每20 min后测定MFI1次,共测定60 min,按照60 min后MFI值不同分为阴性及阳性2组。结果起始MFI值阳性组显著高于阴性组。阴性组MFI随待检测时间增加而逐渐下降,荧光强度衰减与时间成正比,相关关系方程为Y=9.873-0.756X。结论起始荧光强度高于10的样本,在60 min内上机测定不会对阴阳性判断造成影响。起始荧光强度在8-10之间样本,上机测定时间对于阴阳性的判断有一定影响。Objective This study was designed to observe the correlation between the mean fluoresecence intensity(MFI) in HLA-B27 test with flow cytometry and time,to estimate a way to judge the negative and positive of sample by change of time.Methods It is the standard of positive sample that 90% percent of lymphocytes are HLA-B27positive/HLA-B7 negative cells, and MFI is exceed 8. Test 32 positive samples and acquir first MFI, and test the same sample every 20 minutes. Based on the last MFI in 60 minutes, the 32 samples were divided imo two groups: positive and negative group. Results The correlation coegicients with the change of time were 0. 756 for MFI, There was statistically different between positive and negative group for the first MFI. Conclusion the sample of first MFI exceeded 10,the time of testing can not influence the conclusion. But to the the sample of first MFI between 8 to 10, the testing time can influence the conclusion some extem.
